Taking the 1 mL working solution as an example, add 70 μL of 100 mg/ml clarified DMSO stock solution to 400 μL of PEG300, mix evenly to clarify it; add 50 μL of Tween80 to the above system, mix evenly to clarify; then continue to add 480 μL of ddH2O to adjust the volume to 1 mL. The mixed solution should be used immediately for optimal results.

Description
NSC59984 is a p53 pathway activator via induction of mutant p53 protein degradation and p73 activation.

In vitro
NSC59984 specifically restores p53 pathway signalling and induces p73-dependent cell death in mutant p53-expressing human colorectal cancer cells. This compound also induces cell death in a panel of cancer cell lines bearing different p53 mutations with little or no genotoxicity in normal cells.
In Vivo
In nude mice bearing DLD-1 xenograft tumours, NSC59984 (45 mg/kg, i.p.) significantly represses tumour growth in a p73-dependent manner.
Cells are seeded at 5,000 cells per well on 96-well plates. Cells are mixed with an equal volume of CellTiter-Glo reagents, following the manufacturer's protocol, and bioluminescence imaging is measured using the IVIS imager.
